DETERMINING PROTEIN CONCENTRATION
 

Procedure:  We set the Agilent 8543 spectrophotometer to measure the absorbance across a specter of wavelengths from 190nm to 400nm. The spectrophotometer was then blanked using a 500μL cuvette filled with 1X TE buffer. Two samples of purified protein were then measured for absorbance using the above mentioned settings. The output was then analyzed, and manipulated into graphs. The concentrations of the samples were then calculated using Beer’s Law.
