INSERTION AND ISOLATION OF PLASMIDS

Results:

LB/Amp plate with pGlo
   8 colonies grew
   did not glow under UV

LB/Amp/Ara plate with pGlo
   7 colonies grew
   did glow under UV

LB plate without pGlo
   excessive growth
   did glow under UV

LB/Amp plate without pGlo
   no growth
   did not glow under UV

If the cells grew in the presence of amplicillin then we know that the bacteria are expressing the antibiotic gene.  This gene was in the plasmid, therefore, the bacteria absorbed the plasmid and the pGlo gene.

The samples treated with the plasmid on the LB/Amp/Ara plates fluoresced.  This is because they absorbed the plasmid with the pGlo gene.  GFP was then produced from this gene because the gene was turned on by the arabinose in the culture medium.
