Text Box: Prepare Media, Arabinose, and Ampicillin Text Box: Add 250 micro-liters Transformation Solution to each tube (mix) Text Box: Scrape one Colony off plate with loop Text Box: Prepare pGLO Text Box: Streak Plates with E. coli HB 101   Text Box: Label and Pour Plates (& store in refrigerator)

Text Box: Re-hydrate E. coli HB 101

Text Box: Transfer to Microcentrifuge Tube Text Box: Add 250 micro-liters Growth Media (& then incubate for 10 minutes at 37 degrees Celsius) Text Box:  Store at Room Temperature for 2 Nights Text Box: Test plate for Fluorescence with Blacklight Text Box:  Transfer Bacteria to appropriate plates Text Box: Heat Shock in 42 degree Water Bath for 50 seconds, then in Ice Water for 2 minutes Text Box: Add one loop (about 10 micro-liters) pGLO to Tube (& put on ice for 10 minutes)
